Further analysis of the mode of inhibition and activation of Novikoff ascites tumor phosphofructokinase.

چکیده
The Novikoff ascites tumor phosphofructokinase showed a lag in the onset of inhibition by adenosine triphosphate or citrate. Under identical assay conditions, Ehrlich ascites tumor P-fructokinase and the rabbit muscle enzyme were immediately inhibited by ATP. Advantage was taken of this lag period by first incubating the enzyme with ATP for 2 min before the assay of enzyme activity. After the first incubation, the ATP inhibition of tumor P-fructokinase became immediate. This inhibition can be reversed by the addition of orthophosphate, AMP, or fructose-6-P prior to the addition of all the necessary components for assaying enzyme activity. Both the inhibition and the reactivation of the enzyme were found to be dependent on enzyme concentrations. The lower the enzyme concentration, the stronger is the inhibition and the slower the reactivation.

متن کاملPasteur Effect in Novikoff Ascites-hepatoma Cells.
Inhibition of fermentation or of glucose uptake in the presence ofoxygen has been observed in many forms of life and is generally referred to as the 'Pasteur effect'. In most tissues, oxidative inhibition of fermentation (ethanol or lactate production) is reflected by a parallel decrease in glucose consumption. Since certain tissues utilize lactate significantly, estimation of the Pasteur effec...
متن کاملInhibition of glycolysis by amino acids in ascites tumor cells. Specificity and mechanism.
The effect of glutamine and asparagine on glucose metabolism has been studied in ascites tumor cells. Either of these amino acids decreased the glycolytic flux about 80%. Half-maximal effects were obtained with 0.14 mM glutamine and 0.087 mM asparagine. Among the 20 L-amino acids, only glutamate produced a similar effect. Glutamine and asparagine caused a 70% increase of hexose monophosphates a...
متن کاملMatrine inhibits diethylnitrosamine-induced HCC proliferation in rats through inducing apoptosis via p53, Bax-dependent caspase-3 activation pathway and down-regulating MLCK overexpression
The proliferation of hepatocellular carcinoma (HCC) cells is one of the leading causes of liver cancer mortality in humans. The inhibiting effects of matrine on HCC cell proliferation have been studied, but the mechanism of that inhibition has not been fully elucidated. Since, apoptosis plays an important role in HCC cell proliferation. We examined the apoptosis-inducing effect of matrine on tu...
متن کاملCharacterization of nucleolar antigens of normal rat liver, regenerating rat liver, and Novikoff ascites hepatoma cells following in vitro translation of polyadenylic acid-containing messenger RNA.
Nucleolar antigens of normal rat liver, regenerating liver, and Novikoff ascites hepatoma cells were transplanted in vitro from polyadenylic acid-containing messenger RNAs isolated from the respective tissues and immunoprecipitated with specific antinucleolar antibodies and Protein A. By two-dimensional gel electrophoresis of the translation products, five antigens were detected in normal rat l...
